Instech's infusion tethers protect externalized catheters while transmitting torque to a swivel. Instech offers three basic types of tethers: non-surgical harnesses, surgically-implantable buttons, and head block tethers.

Our newest models are the Vascular Access Harness (including dual channel and bile sampling versions) the Vascular Access Button, and the Polyester Felt Mouse Button.

Vascular Access Harness™ (VAH)

 

Instech’s Vascular Access Harness™, an advancement on the Covance Infusion Harness™, permits quick, aseptic connection and disconnection of a catheterized subject and an infusion tether. The system consists of two parts: a miniature external port housed in a harness (VAH95AB) and a mating spring tether with a connector (VAH95T) that pierces the septum to make the fluid connection. Animals can be ordered with a catheter and the harness pre-installed (ask your supplier - or contact us for a list of partners). Then, to begin an infusion study, simply wipe the septum with disinfectant and connect the VAH tether to the harness. Flush or inject directly into the harness as needed using a syringe fitted with an SN22 septum needle or the male VAHLS22/30 connector with needlestick protection.

The standard system is designed for single channel infusion of 100-550g subjects using a 22ga swivel. For best results use the 3Fr polyurethane catheters; a 20cm rounded-tip PU catheter specifically designed for the VAH is available as part of part number VAH95AB-C. The standard harnesses have 9in (23cm) belly bands; use the VAH95AB14 with 14in (36cm) belly bands for 400g+ subjects. All parts are supplied EtO sterilized.

Also available in a two-channel version for simultaneous infusion and sampling or for bile sampling.

Dual Channel Vascular Access Harness™ (VAHD)

 

This new dual channel VAH™ connects two independent fluid channels as simply as the standard VAH connects one. Uses include simultaneous infusion and blood sampling or simultaneous infusion and blood pressure measurement.

Special configurations are available for bile sampling.

Install the VAHD115AB harness at the time that the catheters are implanted. For best results use the 3Fr rounded-tip polyurethane catheters that are included with part number VAHD115AB-C. Access the ports directly using an SN22 needle for manual injections, sampling or flushing. Connect the KVAHD115T tether for continuous access to one channel through a swivel and intermittent access to the second channel through an injection port. Alternatively, connect the VAHD115T tether to a 375/D/22 swivel for continuous access to both channels.

Bile Sampling Harness

 

Instech Solomon's new dual channel VAH™ is ideally suited to bile collection. Install the harness at the same time that 3Fr polyurethane catheters are placed in the bile duct and duodenum. No special occlusion catheters are needed. Attach a loop connector so that bile flow, now diverted through the harness, can resume during transport and rest.

To collect, simply connect a dual VAH tether. Choose one of four configurations (see PDF - or ask for detals) depending on whether you intend to replace the sampled volumes with artificial bile, and whether you prefer tether-mounted collection to minimize mixing and improve time resolution.

The configurations with tether-mounted collection allow for automated bile sampling using Instech's ABS2. The older CIH-based bile reservoir harnesses are still available.

Covance Infusion Harness™

 

Instech's infusion harness combines the best features of jacket and button tethers. Like jacket tethers, the harness does not need to be surgically attached. However, its unique design offers a range of benefits over jacket tethers including adjustability for size/growth, more normal body temperatures and improved air circulation for faster post-operative recovery.

The tether consists of a soft molded elastomer saddle with a vented dome that protects the catheter, adjustable belly bands to secure the saddle to the animal, and standard stainless steel spring stock to protect the externalized catheter and transmit torque to a swivel.

In addition to the single piece tethers listed below, the harness are available in bulk, as replacement parts, or combined with other components in pre-assembled infusion kits. Also available in a configuration for bile collection.

A new variant, the Vascular Access Harness, permits quick and aseptic connection or disconnection of the tether.

Vascular Access Button™

This new implantable button features an external port with a septum similar to the Vascular Access Harness™ for quick, aseptic connection and disconnection of a catheterized subject and an infusion tether.

Compared to the DC95BS, which uses a Dacron® mesh, the VAB95BS buttons feature a sturdier medical-grade polyester felt. Attach a 3Fr polyurethane catheter (included with part number VAB95BS-C) to a 22ga connector under the felt. The button is implanted at the time of catheterization; to begin an infusion study simply disinfect the septum and plug in the mating VAB95T tether. The recessed septum-piercing needle will make the fluid connection without exposing the fluid path to the environment.

Polyester Felt Button for subjects up to 80g

This new implantable button features a 0.563in (14mm) medical-grade polyester felt disk. Catheters are exteriorized through the opening in the stalk, which mates with Instech's standard PS62 tether spring with a press fit.

Dacron Mesh Button Tether

These disposable buttons are designed for infusion studies lasting more than 2 weeks. After one to two weeks the subcutaneous tissue will grow into the Dacron® mesh, making the sutures irrelevant.

The tethers are available in 2 standard sizes (R): .090" clear lumen for single channel infusion and .105" for two channel infusion. (For a button tether for mice, see the LW62.) A complete tether contains one button, 12" spring and a quick-disconnect coupler. The DC95 buttons include a silicone seal for a 3Fr catheter to help reduce catheter movement and bacteria ingress.

Replacement buttons are available in packs of 10 or in bulk quantities of 100 (please contact us for bulk pricing). Implantable buttons are provided individually packaged and EtO sterilized.

Plastic Button Tethers

Instech's plastic button tethers are designed for short- to medium-term infusion studies. All variations of this type of tether are based on the same design: the catheter passes through the stalk of a button which has been surgically attached to the subject (usually under the skin), into a length of stainless steel spring which protects the catheter and transmits torque to the swivel, and up to a swivel mounted outside the cage.

The lightweight plastic will not cause adverse tissue reactions as stainles steel sometimes can. The miniature LW62 for smaller subjects includes Dacron mesh which can be sutured onto the button to expand the attachment area and reduce strain on the sutures.

The buttons have a low profile stalk that reduces stress on the sutures as the animal moves. Simply twist the spring into the button stalk to attach it; the expansion force of the spring will hold it in place.

 

 

LW95/105 tethers also include a quick disconnect coupler (shown above) which sits about one inch above the animal, allowing the tether to be removed and reattached easily. Plastic buttons may be reused but are not autoclavable. They are provided individually packaged and sterilized by ethylene oxide.

Magnetic Head Block Tether

This new design uses magnets to connect the tether to a base that is attached using dental cement. Simply bring the two parts near each other and they will snap together. Tubing exits through a hole in the side of the side of the connector.

Tethers are available either with a protective PS95 spring or a lightweight looped wire. The MMW70 tether includes a special slotted clamp (replacement part MCLAMP) for direct attachment to the outlet tube of our swivels (such as model 375/D/22QM for microdialysis). Use with an MCLA or SMCLA counter-balanced lever arm. The bases are provided sterile; other components are non-sterile.

Head Block Tether (R)

Instech’s head block tether assemblies were designed for microdialysis. Always use a counter-balanced lever arm to remove slack in the head block tether and to give your subject the greatest possible freedom of movement.
 
This large lumen tether can accommodate two standard microdialysis probes. A 3/4" slotted screw attaches with dental cement. A blade on the end of the spring tether slides into the head mount screw and is secured with a knurled tubular nut.

Head Block Tether (M)

Instead of spring, the MINF head block tether uses a fine .010" diameter looped wire, making it extremely light weight and allowing it to transmit torque efficiently to the swivel as the mouse moves. A small peg attaches with dental cement. The wire tether is attached by inserting it into a hole in the peg and sliding a sleeve over it.

The tether includes a special slotted clamp (replacement part MCLAMP) for direct attachment to the outlet tube of our our low-torque single or dual channel swivels (models 375/25 or 375/D/22QM). Always use with an SMCLA counter-balanced lever arm. Use MTUBE collection tube holder for single-channel microdialysis.

Collection Tube Holders

Tether Mounted. The small end of the MCTHQ and MCTHH clips attaches to M115 head block tethers or any tether using PS115 spring stock. The other end holds a standard 1/4 or 1/2 ml collection tube which allows samples to be collected close to the animal and does not require an extra swivel channel for exiting fluid. They are primarily used for multi-probe microdialysis.

Swivel Mounted. The MTUBE collection tube holder is most commonly used for mouse microdialysis. The bracket attaches to the rotating portion of a 375-series swivel using a set screw. A smaller set screw hole is used to attach a MINF or MMW70 head block tether. The bracket holds standard 1/4 ml collection tubes.

Glass Ionomer Cement

This resin-based glass ionomer cement is ideal for permanent head attachment of microdialysis probes and head block tethers. It has significant advantages over the more commonly used methylmethacrylate cements. It bonds to bone, eliminating the need for screws in most cases. It has a lower temperature increase during polymerization. And it hardens more quickly with no noxious fumes.

Our new automixing configuration includes a dispenser, two 13.3gm cartridges and 44 disposable mixing tips. The cartridges have two separate chambers so that the components of the cement are only mixed in the tips; therefore, they do not need to be used all at once. Also, they contain more than 80 times the volume of the 0.16gm single use capsules that we previously offered. An SOP for rodent head attachment is included. Sold for research applications only (not for clinical use).
